The LS-5B was generously donated by Monsanto in St. Louis. It is currently used in both CHEM 322, Instrumental analysis and in undergraduate research projects in chemistry. The instrument has scanning excitation and emission monochromators and is capable of both fluorescence and phosphorescence measurements.
Using the LS-5B:
The LS-5B is controlled using the keypad on the instrument. Data is displayed digitally on the display panel of the instrument, and can be printed using the associated printer. We hope to interface the instrument to a computer in the near future for data collection and storage. Below is an image map of the instrument control panel. Click on a letter to see a description of the associated keys.
A: Experiment Selection Keys back
Allow you to choose fluorescence (FLUOR) or phosphorescence (PHOS) mode
B: Numeric Keypad back
Pretty self explanatory. If you need to enter a number, this is the thing to use!
C: Monchromator Controls back
This collection of four keys, combined with the numeric keypad, allow you to position the excitation and emission mochromators at the wavelength of your choosing, and select the wavelength range of the monochromator that is to be scanned (if needed). The left GOTO l button sends the excitation monchromator to the wavelength displayed under the "Excitation" heading in the display. The right GOTO l button does the same, except for the emission monchromator.
The LOW l and HIGH l buttons set the upper and lower wavelengths for the monochromator that has been selected using the Monochromator Selection Keys, described in part D.
D: Monochromator Selection Keys back
Allow selection of the excitation (EX), emission (EM), or both (EX/EM) monochromators for control the rest of the control panel.
E: Monochromator Scan Speed back
Allows variation of the motor speed of the scanning monochromator through 5 preset values.
F: Manual Monochromator Drive back
Allows fast (<< and >>) and slow (>, 1 nm increment) manual control of the monochromator selected using the Monochromator Selection Keys.
G: Start/Stop back
Begins and manually terminates a scan of the monochromator that is selected with the Monochromator Selection Keys.
